QuickChange mutagenesis: how to do it without kit? - What dNTP concentration should I use? (Dec/19/2005 )

Hi, all,

I want to do a mutagenesis in my plasmid, using the methodology of Quich Chanege Kit by Stratagene - PCR, after DpnI restriction, etc.

Their secret is the concentration of nucleotides dNTP to perform the PCR ethap. Maybe somebody do it already by himself, and can suggest me a dNTP concentration, which I should use in this reaction.

I use 200 µM each and it works fine. As long as you have excess, you should be ok.
