yeast transformation question... - (Nov/21/2001 )

use about 1ug of the crude DNA, that will do. If you use too much then efficiency starts dropping.

cheers

Hi

I'm transforming yeast using a standard LiAc/PEG method (if there is such a thing!). Does anyone have information on the purity of DNA needed for such a transformation...is a crude ispropanol pptn from lysed e.coli OK, or is an actual mini/midiprep needed. Also, is there a maximum amount of DNA you can use in a transfomation or is it a case of more the merrier?

Cheers

Chris
