Sticky end PCR - Attractive method... (Dec/07/2005 )

Does anyone has any experience using sticky end PCR (Zeng,G. (1998) Sticky-end PCR: new method for subcloning, Biotechniques)? Since I´m going crazy with my (failed) attempts of cloning, I thought of giving it a try. Thanks!!

I would personally try cloning into a TOPO vector first before using that method.

Yes, that method is cute, but it doesn't tell you if the ends are "stickied" properly.

I mean, TOPO is automatic, pretty much...
