Minipreps from Genehogs cell line - (Dec/05/2005 )

I am trying to get a clean miniprep from Genehogs (derived from DH10B) cell line. Everytime the minipreps look fine but produce smears when digested with REs (looks like a nuclease problem - is this possible with this cell line?) The supernatant obtained after the Potassium acetate step looks milky.

DH10B is endA-, so it has the important endonuclease knockout of plasmid prep strains. I'd suggest that you reduce the size of your pellet in the prep. Milky supernatant usually indicates that you have overloaded the prep with too many cells.
