HindIII digestion - (Dec/03/2005 )

Hi all,

I'd like to know how many units HindIII I should use to digest 12ug BAC DNA overnight. I know 1U is required to digest 1ug DNA but I have found conflicting info about that so I'm a little confused...

20 units will do the job. Generally i use 1µl in 50µl reation to limit the glycerol final conc.
