How to effectively recover 40 bp-long DNA fragment from restricted reaction? I need your suggestion! Thank you!

Perhaps you could use high percentage agarose (say 3-4%). Or native TBE/polyacrylamide gel that you stain after with EtBr..
A cheap alternative would be ordering complementary 40 nucl oligos, provided you know the sequence (if not, have it sequenced).
