Actinomycin D is usually given i.v. and can cause severe local necrosis if extravasation occurs. Therefore, it should be administered into the tubing of a rapidly flowing i.v. infusion. The Plasma half-life i s very short, but tissue half-life is long (48 hrs) due to DNA binding. Only a small amount of the drug is known to be metabolized. Urinary and fecal excretion of the drug is prolonged. In humans it does not appear to enter into the central nervous system .
