What happens if I make a ligation more than 16 hours?

At which temperature?

Blunt or sticky ends?

At 16C I generally do for 24 hrs(during weekends ).. works better for me...

It doesn't matter over 16h and may has some good effect on your ligation efficiency.

I made the experience, that people who ligate over night just want to go home
