ACE PCR - problems with PCR for angiotensin-converting enzyme (Nov/25/2005 )

Hi!
I've got problems with PCR for angiotensin-converting enzyme polymorphism. This polymorphism concerns small deletion beetwen exon and intro. My PCR only shows results for homozygotes. It's troubble with heterozygotes. PCR product has about 600 bp, PCR product with deletion has about 300 bp lenght. Is it maybe polymerase preferences? There are problems with longer product in heterozygote sample.
Maybe you know where is the problem and you can help me resolve it.

Hello,

i remember that some of my friend was workinkon ACE and have this kind of problem, she make two different PCR to be soure tere is no false results. There is indeed a preferentially amplified allele. Maybe it was a nested PCR she was making but i can't remember i think there is an article but i'm not soure.

i forgot, there is some Taqman SNP Genotyping Assays for ACE which work very well if you have the material.
