Streptavidin-Alexa fluor 488 - it should be dissolved in PBS or PBS/BSA? - What concentration of it do you use, and how long is incubation? (Nov/23/2005 )

Hi,

I try to establishe the conditions of immunofluorescence test.

Unfortunately on the datasheeet of straptavidin-Alexa 488 conjugate they do not give the information, in which solution it should work, and at what concentration.

So i want to gather some data from the experienced guys:

In which solution better to incubate streptavidine - Alexa 488, in PBS only or in PBS/BSA?

What shold be (about) its concentration. I know, that the gamme of dilution should be done, etc, but I have not a lot of time and, moreover, I should have at least any idea, from what value I should begin a gamme?

First and foremost, CALL TECHNICAL SERVICE and ask them how it should be diluted. Many antibodies just need dH20 as they are actually lyophilized with buffer components already. Call and ask though to be sure. In any case make sure whatever you use is sterile.

As far as concentration, I use my Strep-Alexa 488 at 1-2 ug/ml which for my vial is about 1:1000.

Don't add BSA. It will be fine in PBS. The avidity between streptavidin and biotin is huge. They bind perfectly. Plus, why add extra protein, it's not needed.
