ATP hydrolysis - (Feb/09/2001 )

We are doing some ATP dependent enzyme assays. As a control for nonspecific reaction we substitute ATP with AMP. Unfortunately, we get a substantial amount of reaction going even with AMP, which we suspect that it is due to ATP contamination of the AMP. Does anybody have an easy method to deplete the ATP from the AMP, e.g.enzymatically with maybe a phosphatase? I would rather not go through the process of doing an HPLC purification. Or does anyone know of a source of AMP that is free of ATP?

you may want to treat with adenosine 5'triphosphatase (atpase) enzyme specifically for digesting atp to adp. common source of amp is from yeast. generally, amp derived from yeast is free of atp. from experience i've used amp from sigma chemical and have not found any batches that had any atp contamination.
