How to use MG132? - (Nov/12/2005 )

Hi, everyone:
In my experiment, I need to use MG132( a kind of proteasome inhibitor). I read some papers which all just provid the concerntration used on the cells, such as 10uM,but I still don't know the total amount used on the cells. For example, I treat the cells in the 10cm dish, how much I should use? Does anyone can help me? In addition, whether I should treat MG132 in SF medium or complete medium?? what the maximum hours I can use it on the cells??

well, 10uM is the final concentration of the inhibitor in your medium. calculate the concentration of your stock solution and dilute to final 10uM in your medium.
type of medium would depend on type of the experiment you want to do.
for the lenght - people use different times of exposure.. depends on the experiment you want to do. you have to remember, that this stuff is a bit toxic, so too long is not good.
