tris-tricine gel transfer - (Nov/08/2005 )

Hello everybody!

Does anyone have any experience with tris/tricine gel transfer? I'm trying to separate 6kDa protein using ready Tris-tricine precast gel system of Bio-Rad. Afterwards I transfer the proteins into Protran nitrocellulose transfer membranes for 1 hour. Is there any optimal transfer time? Or may be it's too long? Is 1 hour long enough because I don't see much of my protein on the membrane.

1 hour is not too long i think.
You can increase time at same voltage, or increase step by step the voltage for transfert, for same time
For me, i would play with time, as some electrodes may be affected by high voltages... (22 or more for mine..)

6 kDa is small

I have a little experience with small proteins and it is easy to lose them

I would switch to PVDF, 0.2um pore size
