DNA treatments - PDNA chloroform treatment (Nov/07/2005 )

hi all what happens to plasmid DNA when it is treated with solvents like chloroform or methylene chloride?will it denature it?Mostly DNA will be in TE buffer , emulsified with chloroform?

i think that not goes very wrong for DNA in phenol treatment during a purification, as it hols an aqueous area near him...
But as silicas based extraction kits are more and more developped, i think it's maybe better for DNA (but major point of them is the fact that it's quicker and less toxic for you and the environment)

thanks.I am not using solvents for DNA extraction.This is some what polymer chemistry related stuff. I have another doubt in general chloroform will denature proteins. I am using Polyethlenimine which is a polyamine.It has got primary , secondary and tertiary amino groups. I am using it as a non viral gene vector.I will couple it with DNA .The DNA will bind with the primary amine group.So i have the following doubts,
1.PEI is hydrophilic.But whan I dissolve it in chloroform , it did dissolve.So what happens to the structure of PEI in solvents.
2.I want to know what happens to it when treated with solvent as such.
3.When coupled with DNA ,treating it with solvents,what likely may occur? will the solvent denature the complexes?
