how to isolate plasmid insert? - (Oct/24/2005 )

i have a brain twisting question. i was given a plasmid with an 5kb DNA insert. what would be the best and simple method for me to isolate this insert. a step to step method would be appreciated. note, i don't know the sequence of the insert. all i know is this insert is next to a universal priming site and the insert is about 5kb.

look up the map to the plasmid, it'll have all the restricion sites. hopefully your insert is within the MCS, then you can do an easy restriction digest. anyway, look at the map.
