Question about bacterial growth - (Oct/24/2005 )

Does anybody know, how long it takes bacteria growth to reach OD=0.6 from single colony inoculation. basically, 6-8 hr? Well, if you got transformation plate from e.g BL21 (DE3) and you inoculate one colony to 50 ml LB with antibiotic in 37 degree 250 rpm shaker, how long it will reach OD (600nm)=0.6? Thanks.

you can in first option take the OD at starts point. Then, number of bacterias (E. coli) will double every 20'. So you can calculate the time you'll have OD=0/6.
Second: seed the culture and take OD after half an hour. Then same as above mentionned. The predicted time will be more accurate.
For my comp cells, i start a 10ml preculture and seed 1l culture. takes about 3h to reach 0.6

Hi

I usually set up an O/N of a single colony into 5/10mls & then next day subcultures 100x dilution into fresh media. It then takes approx 2-3 hours to reach 0.6 OD600.
