Tris-buffered phenol:chloroform:isoamyl alcohol - How do I prepare? (Oct/14/2005 )

Hi,

I just received phenol:chloroform:isoamyl alcohol (PCIA) with the Tris in a seperate bottle. How do I mix them to make tris-buffered PCIA?

normally, pre mixed P/C/IAA are prepared with saturated phenol. Check it with the manufacturer. Moreover, you should take care of the pH of the final solution.
For DNA extraction, pH is 8
for RNA extraction pH is 4.7
