SDS-PAGE and western blot - (Oct/12/2005 )

Please answer my question.

I just wondering if it is possible to boil the samples with the loading buffer for the SDS-PAGE, then stored them in the -20C. When I need them, I just thaw them and load them onto the SDS-PAGE.

Will the samples be OK? Will the western blot be fine?

everything will be fine. when you take them out from -20 you can worm them up to 70 if you want.

On average for a simple Western blot your protein should be fine. However, every protein is different..and unfortunately some things, esp when it comes to proteins are totally empirical.

However I did read of a guy that had very rare samples, and he stored as you would like for over 10yrs and each time Westerns worked fine. Personally I store the samples w/out sample buffer.
But I have done both w/no ill effects.
