scale-up Dnase reaction - is it possible (Oct/04/2005 )

Hi there,

I would like to know if it is possible to scale-up a Dnase I reaction. It means: I am treating RNA extracted with Trizol, if the protocol says to make a 10 ul reaction containing 1 ug or RNA and 1U of enzyme, can I make 100 ul or even 200 ul reaction and add the proportional amounts of RNA(10, 20 ug) and enzyme (10, 20 U)? or do I have to make 10 or 20 different small reactions in 10 or 20 tubes?
Does the scaling up affect in any way the reaction?

Hi,
You can definetely scale up the reaction, it will work the same as before, infact i've done it several times successfully, and when trying with different no. of samples, try making master mix.
