How well do macrophages respond to DMSO? - (Nov/22/2009 )

I am culturing J774.2 macrophages for the purposes of exposing them to amyloid beta and determining how that affects tnf-a.
i am using several potential inhibitors and I plan to dilute them in DMSO. I know that the ideal is below 0.5% dmso diluted with the culture medium, but are macrophages less sensitive to it or more sensitive?
Also, I am culturing outside of a co2 incubator so my medium contains HEPES buffer.
Should I dilute the DMSO with the medium containing HEPES, without HEPES, or does it not really matter?
