Sorry the rating actually does not really apply because I have not tried the technique yet. I have never done any dot blots but would want to attempt some (also for antibody concentration optimisation) if I had known how to incubate on one sheet with different concentrations antibody ? Do you apply eg. a few microliters/1ml? of each antibody concentration only to the blot on each block (I would expect it to spread and contaminate each other) or do you cut your large square into small squares and incubate them separately in separate smaller containers? Otherwise how do you prevent contamination between samples on the different blocks, adjacent to each other?
Please clear up the mystery for a "non- molecular biologist"!
