desulphonation on column - (Aug/23/2005 )

Has somebody ever tried to desulphonate the treated DNA directly on the (silica)column like in the Chemicon kit? I just wonder why to elute, desulphonate and precipitate again.

you need to denature the DNA in order to desulfonate it. DNA, in double stranded form will bind to the silica, single stranded would bind much less efficently.
