PCR primers for Mycoplasma detection? - Publication gives many, which to choose? (Aug/23/2005 )

I need a sequences of Mycoplasma PCR detection oligos, I have found many oligos sequences in publication of

Uphoff CC Drexler HG, Detection of Mycoplasma Contamination, from: Methods in Molecular Biology, vol. 290: Basic Cell Culture protocols, Third edition.Humana pressInc., Totowa, NJ

And I just wonder, why they give six sequences for 5' primers, and 3 sequences for 3' primers.

Maybe somebody could write me just one working 5' and one 3' sequence to perform such a PCR in a non-complicated way?

Hi Alesia,

I suppose the different primers are for the different mycoplasma species. I've successfully used the following primer sequence for mycoplasma to detect the whole genus:

For: 5' GGG AGC AAA CAG GAT TAG ATA CCC T 3'
Rev: 5' TGC ACC ATC TGT CAC TCT GTT AAC CTC 3'
Product: 280 bp

They were pulled out from Vojdani et al (1999): Multiplex PCR for the Detection of Mycoplasma fermentans, M. hominism, and M. penetrans in Patients with Chronic Fatigue Syndrom, Fibromyalgia, Rheumatoid Arthritis, and Gulf War Syndrome

However, as I don't work in the area of mycoplasma detection, I won't be able to tell you if these primers are THE best. Read the paper, and maybe you'll be better informed whether you should use the sequence. Good luck!
