reporter assays - luciferase reporter assay (Aug/22/2005 )

i had transfected hela cells with luciferase plasmid (1ug) having sv40 promoter but m not getting any luciferase activity after transfection. i had checked transfection efficiency by co-transfecting GFP sad.gif please help!!

First of all, you should check the information of the cell line you used in promoter assay from other papers. Maybe the SV40 promoter in your cells would not be driven well. The second thing you have to notice is that anyone has used this construct in promoter assay(even in the same tube)and got a successful data in your lab. I mean maybe the SV40 promoter has some mutation problem in this construct which I have ever encountered.

thanks for ur suggestion......i had compared luciferase activity with another SV40 construct and it had given me 3 fold better activity as compared to what i was using.
