How to quantitate digested DNA by spectrophotometry - (Aug/22/2005 )

Is there some trick to getting the concentration of digested genomic DNA through spectrophotometry?
I am digesting with DpnII and the gel picture suggests that there is DNA there, but the spec is saying there is nothing there.

Also, what would the gel picture of DpnII digestion look like?

There aren't any tricks to getting the concentration via spec, though you may have to purify the DNA before the spec will give the right reading.

I would suggest having a look at the NEBcutter website (New England Biolabs website) to make a virtual gel of your digest.
