Unmethylated control - (Aug/08/2005 )

Hy everyone!

I have read in some literature that it is used sperm DNA like unmethylated control, and SssI-treated sperm DNA like methylated control. My question is: Does someone use this kind of controls? If the answer is "yes", There is a comercial product of it? or Which is the protocol to obtain the sperm DNA from the biological sample?

hi, I want to perform methylight,I haven't found a comericial product of sperm DNA, and in some literature I find human WBC DNA was used as unmethylated control.

Thank you for your reply JWD. It could be very interesting, but I am sorry because my English is very bad, but I do not know what is human WBC DNA. Can you explain me what is it please?

Ok, I am now here. I know what is WBC DNA (white blood cells DNA). But WBC DNA has a standard methylation pattern and probably there will be genes that are methylated or hemimethylated, so it is no a good unmethylated control.
I was using that for unmethylated control, but there is one gene that is hemimethylated in that control samples. That is the reason that I want a total unmethylated control.
