Hi Nick,

Thank you for you tell me in using longer PCR primer. So I think the more longer the primer, the less like using nested primer. Is this conception oK?

not necessarliy,

the longer the primer leads to increased specificity of the primer, and with bisulfite converted DNA this is essential as the complexity of DNA decreases.

You still however would require a nested primer to ensure you will amplify enough template to sequence, clone or restrict.
