Does Temperature Alter DNA Methylation Status - Temperature and Methylation (Jul/25/2005 )

Hello All,

I was doing some searching on the web for papers disscussing temperature effects on methylated DNA.

Specifically I was wondering if I was changing the DNA methylation status during DNA extraction (done at room temperature) from human tissue.

I'm considering an experiment where I would fully methylate genomic DNA using M. Sss I, with various temperature storage conditions. Then I would cut with McrBC restriction endonuclease.

Has anyone performed such an experiment and willing to share results? Or guide me towards work done in answering this small technical question?
