Growing prostate carcinoma PC-3 - (Jun/29/2005 )

Hi,

I thawed some PC-3 cells that I had received fron ATCC 2 weeks ago. The cells have been fed using the ATCC suggested media (Kaighn'modified Hank's F12 w/ 10%FBS) but they do not seem to grow. Less than 10% of the cells differenciate. There is no contamination of my media.

Are there any vitamins that can be added to the media to boost the cells? Should I consider reseeding them at a higher density?

Hi

I am growing PC3 cells in DMEM/F12 medium (Gibco) with 10% FBS. They are growing fine.

btw, in case you didn't know, ATCC will give you a credit if the cells die within 30 days of purchase.

We grow PC-3 cells in RPMI medium with 10% FBS and PS. No problem at all.

I discussed my cell growth problems with a tech at ATCC and I think myproblems are linked to the serum we use. We heat-inactivate FBS before use. ATCC suggested this could degrade valuable proteins/hormones. Did you observe the same thing with PC-3 or OVCAR-3?
