ChIP with primary human CD4+ Lymphocytes possible ? - (May/26/2005 )

Hello all,

I'd like to know if someone tried to make Chromatin immunoprecipitation experiments with lymphocytes. Is there a way of optimizing conditions? Because it seems there are not enough CD4+ lymphocytes in a buffy coat to complete a full experiment.

Depends how many you can obtain from each buffy coat. A common ChIP procedure many people use requires about one million cells.

Depending on how active your transcription factor of interest is, you may be able to get away with using fewer cells. I would assume fewer cells could be used if you were looking at histones as well. There's only one way to find out.
