taqman probe design for MSP - (Apr/29/2005 )

hello all, i'm have a new project in dna methylation. have limited prev. experience of primer express. project involves Q-MSP with taqman primers/probe. i am told that ensembl and methprimer r 2 v. good programs for methylation...etc. i take it you can't use primer express to design methylation specific primers so if i use meth primer how do i then design the taqman probe??

cheers if you can help

Hi Dave,

this message should be moved into the DNA methylation Forum.

Have you had a look at the topics found in this forum?

My suggestion to design taqman probes is to "mock" bisulfite convert your seqeunce before entering it into primer express. That is to say change all your C's to T's and leave your CG's as CG's (assuming your promoter is methylated). Hopefully primer express would be able to select a suitable taqman probe for you.

Good luck
