5 Aza Treatment Length - (Apr/28/2005 )

Hi,

I had a quick question about demethylating using 5 aza treatment on breast cancer cells (in culture). I've seen recommendations for 5 day treatment and 8 day treatments. What is the minimum treatment period required for full demethylation of the DNA?

Also, my understanding is that 5 aza is extremely unstable in an aqueous solution. Is it necessary to refeed every few hours? or will every 2-3 days suffice?

5azadC treatment is cell specific. Thus you will need to optimise the level of 5azadC for your particular cell line. Be warned, some cells are resistant to the drug if they have active cytidine deaminase. The drug has a short half life, with my experiments I refresh the drug in the media daily.
