Multiplex MSP? - (Apr/07/2005 )

Hi, I'm Elena and I'm working in methylation using MSP. I had optimized MSP for different genes, but it could be more quicker if I could test two or more genes together. I'm wondering if is possible to optimize a MSP for different pairs of primers, like a master mix PCR, and I want to know if anyone know about it or have read some paper using something like this.

Do you mean multiplex PCR? It is possible but much harder than regular PCR.

I agree with pcrman,

you would have to optimise each primer set individually before multiplexing.

If you will be doing this on a regular basis then it's worth the trouble.

If you are truly intersted in multiplexing pcr reactions, you need to start from the ground up, and design your primers in such a way that they all work under the same conditions. If primers won't work using the pre-determined conditions, make new primers.

This way, when it's time to multiplex, all (or most of) the hard work is done.
