USING MISONIX MICROSON FOR SONICATION FOR CHIP ANALYSIS - (Apr/01/2005 )

DOES ANYBODY IN THIS FORUM USE MICROSON SONICATOR FROM MISONIX FOR SONICATION FOR CHIP ANALYSIS. IF YES, COULD YOU PLEASE GIVE ME THE CONDITIONS THAT YOU USE AND IT IS WORKING....

Other's conditions may not work for you. It is very important to obtain optimal conditions by running preliminary experiments.

I use the microson misonix sonicator for my chip experiments. I always get DNA of 200-1000bp size. i use it at a setting in between 2 & 3, something like a 2.5
I sonicate the samples for 10 times with 10 sec pulses. At each 10sec interval rest the samples on ice for 1min.

Even using the same sonicator, your conditions WILL differ depending on the cell type used, the extent of crosslinking, the amount of cells you're sonicating, the volume you're sonicating in, and the tube you're using. It's best to optimize first.
