"Cell culture discard in chloros (10,000) ppm" ? - (Mar/23/2005 )

It says; "Cell culture discard in chloros (10,000) ppm must be kept in the cabinet for a minimum of two hours (preferably overnight) prior to discarding down the sink with copious amounts of water ". What does it mean with "Cell culture discard in chloros (10,000) ppm" ? And would not this be very riskable for contamination if i let the cell culture in the cabinet before throwing it?

Hi there,

I think that the amount of chloros that you add to disinfect your cells in 10,000 parts per million. One ppm is 1 part in 1,000,000. The common unit mg/liter is equal to ppm. So for example four drops of ink in a 55-gallon barrel of water would produce an "ink concentration" of 1 ppm.

Personally, to disinfect cells I use Virkon diluted down roughly to 1/20 and then let the beaker with the waste sit in the sink for about 1 hour, after that add lots of water and pour it down the sink! I wouldn't leave your waste to stand in the hood. Only do that, if you are in the hood and are getting rid of more cells etc...

Should we always disinfect cells first and then spill it down with water when we throw the cultures? Would not this be too risky, since the medium will be spread to the air for the microorganisms?

Are there any differences if the cultures are contaminated or not?
