isolation of mitochondria from mouse fibroblasts - (Nov/26/2004 )
hi,
Can anyone help me to find a protocol for isolation of mitochondria from mouse fibroblasts?
I need to recover an huge quantity since I must do a ketoglutaric dehydrogenase assay on WT and knock out mice , but I isolate solely few mitochondria from 50 x 10^7 using a mitochodria isolation Kit (Pierce).....
the efficiency of Kit is low, and I read, in a paper of biochemistry, that some authors
use about 3x10^8 cells to obtain 3-4 mg/ml of mitochondria....
