can't detect bcl-2 in cytosol - but Bax is there! (Dec/15/2008 )

I did cellular fractionation and managed to detect Bax in the cytosol of my samples, but Bcl-2 is not there. in fact Bcl-2 is in my nuclei fraction. why is that? isn't bcl-2 supposed to be in cytosol?

The protein product of the oncogene bcl-2 is a component of the nuclear envelope, the endoplasmic reticulum, and the outer mitochondrial membrane.

Hope this helps!

thanks, my supervisor told me the same thing.

I thought some of it floats in cytosol like Bax. I was wrong
