Mixed disulfides with glutathione - (Dec/13/2008 )

Hello everyone,

Has anyone ever tried creating a glutathione mixed disulfide with a protein (GS-S-protein)? Would this reaction happen spontaneously?

what exactly u want t o do (Question not clear)

Glutathione will create disulfide bonds in the refolding mix .

I would like to create a mixed disulfide of glutathione with a protein that I am studying. What are steps that I can do to cause this to happen stably.

Would I incubate oxidized glutathione with the protein, or would reduced glutathione work? My one worry working with oxidized glutathione is that is may cause crosslinking of the protein on the cysteine residues.
