Mioblasts primary cell culture - counting the number of myoblasts (Nov/26/2008 )

Hi everybody!!
I have recently started with the culture of mouse myoblasts and have lots of doubts but is not easy to find practice info about this cultures. The main problem I am having is how to identify them inmediatley after isolation because i need to seed these primary extract from two types of mice at the same density and i am not able to distinguish cells from debrils at the begining. Any practical staining? or advices to identify them??If anybody could show me a picture of these primary myoblasts inmediately after isolation it would be perfect.

Hi Rachel,

What you need to do is count the cells using hemacytometer immediately following isolation. The cells would be round and bright, very different from debris. that way you can afterwards culturte them at the density you want.

by the way, i don't know what is your methos for isolating these cells, but consider the fact that in the muscle tissue exist also other types of cells, which are not myoblasts (adipocytes, blood cells of all sorts, fibroblasts and more).
