ELISA - Capturing and detecting Abs concentration - (Nov/24/2008 )

What would be the ideal concentration to use for capturing and detecting antibodies? From my understanding, capturing Ab would be at least 1ug/ml and usually much more higher than detecting Ab, which is usually prepared in much lower concentration. Please advice, thanks...

The ideal concentration must be determend for the specific test by checkerboardtitrations.
Capturing with 1- 10 ug/ml is a good starting point but for the detecting antibody it is dependend form the quality of the antibody en the detectionsystem used Enzym,Fluormetric etc. from 1:1000 up to 1:100,000 and maybe higher
