WB of beta amyloid - a simple method for WB to visualize b-amyloid? (Nov/10/2008 )

Hi,
I am new in beta-amyloid field.. I am interested in running a SDS gel to visualize b-amyloid. I have found several methods, but I cannot make many trials as I dont have many samples. Do you think a 16.5% of Tris-Glycine, 120 V, is enough?
Do I have to take care about boiling, SDS, beta-mercaptoethanol...I bought a color marker to avoid running off my beta-amyloid.

you should use tris-tricine.

you do need to use sds, 2-me. a-beta is "sticky", you need to ensure that the peptide remains a monomer.

I attach a very good paper about Tricine gels.
I have a Bio-Rad Protean II Unit for electrophoresis (the most common, I guess) Once I have mounted the gels..do I have to put the cathode buffer inside the basket with the gels? (so the anode will be outside)

the cathode buffer goes into the buffer well that covers the gel's teeth (ie where you load the sample).

the proteins will travel from the cathode to the anode.
