How to prepare buffers - (Oct/29/2008 )

I need a few hints on how to prepare a phosphate buffer

Details

Total phosphate concentration = 0.025 mol/L
pH =3

Strange buffer, never seen
Take 0.025 M/l fosforous acid and adjust the pH with NaOH 0.1 M

Why Strange? You have never worked with HPLC I assumed. Ever heard of HH equation

pH=pKa +Log [A-]/[HA]

where [A-] +[HA] =0.025 ?

It is strange because in real research, we seldom use phosphate buffer at pH3.0.
