total protein extraction from brain tissue - (Oct/13/2008 )

Hey everybody!!!


I need to extract total protein from frozen brain tissue, for homogenization i have a mortar and pestle.
We thought to grind the tissue manually in mortar while it is in lysis buffer (the same one used for extraction from cells).

Does anyone have any suggestions what i should do , also i'm surching for more organized protocol for this matter .
If anyone have protocol for extraction T. protein from brain or have a link to it please smile.gif please make my day and send me.

just don't forget protease inhibitors

try to grind under liquid nitrogen: this is a protocol to extract instabile metabolites such as ATP but should also be good for protein extraction

we homogenize thawed brain tissue in a teflon/glass homogenizer with buffer then spin away the insoluble material.
