antibodies working in indirect ELISA but not in inhibition ELISA - (Oct/04/2008 )

I really couldn't tell you. Antibodies work in mysterious ways... =)

But can't you do the inhibition ELISA with a sandwich in the end, rather than a direct ELISA? Sure, you need an extra incubation for coating the wells and another set of antibodies, but it should work. Right?

yes, what I learnt is that abs are really strange, sometimes. Yes I'll develop a sandwich ELISA, but I need to use the monoclonals as presenting abs. It means I have to purify them (and it will take much more time with respect to polyclonals) and hope they will work, because this possibility has not been tested, yet. Wishing well..
