Transfection in macrophage - It is possible?? (Sep/24/2008 )

Hello everybody,

Could anybody recommend me a protocol for transfection in primary murine macrophages or in J774.A1 cell line , or give me any piece of advise for it?, because I was asking in my lab but they tell me that is not possible.

Really, Is it possible or not?, lipofectamine or electroporation? would be better try th transfection in another cell line like RAW?

Hello, Amaxa nucleofector can trasfect every kind of cell but you must have their electroporator and kits (expensive). Or you can see this : http://www.ncbi.nlm.nih.gov/pubmed/11058814 and this: http://www.polyplus-transfection.com/EN/pr...t.php?PAGEID=23.

Amaxa would have some success with the RAW cell line - as they are deficient in the sensor of cytoplasmic dna tht most otehr macrophage lines have. That's why transfection is so hard, they respond to it.
Your primary lines would be near impossible to transfect - try a retorvirus - or maybe the amaxa. Very limited success would result from lipid-based transfection reagents.
