PVDF membrane - western blot (Sep/23/2008 )

hi everyone

I did western blot sometimes, everytime before i was ready to transfer protein to PVDF membrane, I must dip the membrane in the methanol first.

could you tell me why do i do it before transfer? what is the principle? thanks in advance!

to hydrate (wet) your membrane

only to hydrate the membrane? any other reasons?

Hi!
PVDF membranes are extremely hydrophobic and usually will not wet in aqueous solutions unless pre-wetted with methanol.
