pH in Medium - RPMI (Sep/12/2008 )

Hello,

I need something else to buffer the pH in RPMI. I already used the maximal dose for my THP-1 cells but the medium ist still changing the pH upon treatment with my drug of intrest.
Is there anything else I can use? I want to avoid medium change in the middle of the incubation time because the half-life os my drug is relatively short and i would have to add new one to the new medium but this would change the whole experiment (I am looking for transcription factors).

Any suggests?

I had a similar problem of medium changing color to yellowish after addition of NAC. I have contacted authors of many papers who used it and they all replied that it should be neutralized immediately before addition to the culture medium.

We use RPMI-1640 medium that is additionally buffered with 25mM HEPES. This can be bought commersially. I have looked into the litterature HEPES should not harm the cells.

HEPES I already tried. I now will do some experiments to determine the influence of medium changing in the middle of the incubation time. Fortunately my drug of intrest is also visible in FACS so I can just check how much cells get destained by just adding two more ml of medium.
