Bioluminescent bacteria - (Sep/12/2008 )

I am trying to freeze dry bioluminescent bacteria in 96 well microtitre plates. Although I am seeing a good post-freeze drying bioluminescence, I am getting huge well-well and column-column variation in bioluminescence. I am using homogenous (I believe) cell suspension, not able to understand what is the cause for this variation (I am using the same bacteria in every column and row. Any inputs or suggestions much appreciated.

TO me bioluminescence is a tricky business. I have been working with luminescent=based biosensor and most of the time you will get variation . even though u repeat it under the " same condition" . luminescent involved a lot of genes ( lux CDABE).. . maybe gene expression got altered , oxygen content , agitation .. all these factors contribute to the inconsistency.
