Uhm.....turns out when I send my primer order to the TA i didnt specify 5-3 or 3-5 so she assumed that my complementary sequence was 5-3, which it wasnt. To me it was quite obvious if you looked at the sequences because I had TTTTTT in one and AAAAAA in the other one but apparently its not.
This is the worst start in a PHd EVER, i look like an idiot, i feel like one, and im getting acne lol
When ordering primers by default they are done 5'-3' unless you notify, could be obvious for you but for the others is just a sequence of nucleotides and hard to tell what is sense or antisense.
Agree with merlav. Also, people who take your orders are not suppose to guess what you want. I certainly would't want a tech to change the orientation of my primers because he/she thought it was messed up.
Now everyone makes mistake. Give yourself time !
Yeah i told the boss and she took it well, I mean Ireally didnt know about this default 5-3 law,oh well! at least im going to have awesome kick ass annealed oligos
