could somebody can give me a protocol of Gus staining? especially for the solution.thanks in advance

hi, i do not know what kind of protocol you want, i usually just transfer the material into GUS buffer, incubate at 37 degree overnight. Next day, examine the transformation efficiency under microscope. some times you have to wash your materla before put them in, and if you use color materla like leaves, you have to decolor them first before you can the the GUS effect.

GUS buffer

50mM Tris.HCl (pH 7.0)
125mM CaCl2.H2O
0.5mM K3(Fe(CN)6)
0.5mM K4(Fe(CN)6)
10% Triton-X-100
pH 7.0
add 30mg/40ml X-Gluc before use

first i must thanks very much for your help. i think it is very useful. some days ago i did the GUS staining with the buffer without K3(Fe(CN)6 and K4(Fe(CN)6.
And sometimes i did the GUS staining overnight but the signal is spreading I do not why? my promoter only express in several cells in the root but the GUS signal spreaded to many cells around the aim.
